Molecular overview
KPV is the tripeptide lysine–proline–valine, corresponding to the C-terminal residues 11–13 of alpha-melanocyte-stimulating hormone (α-MSH). Its small size distinguishes it from the full 13-residue hormone and from larger melanocortin analogues.

In intestinal epithelial cell models, KPV uptake has been linked to the di/tripeptide transporter PepT1, with reported reductions in NF-κB and MAP-kinase inflammatory signaling and pro-inflammatory cytokine expression. Whether KPV acts through melanocortin receptors, intracellularly after transport, or both, has not been fully resolved.
Published-research summary
Key studies used chemically induced colitis models in mice, reporting reduced inflammatory markers and histological changes after KPV exposure. These are a limited number of preclinical studies from a small set of research groups.
Rodent colitis models do not establish human efficacy or safety, and no therapeutic conclusion should be drawn from them.
