Reconstitution Chemistry for Research Peptides

Diluent choice, concentration maths and technique. The three things that determine whether your solution matches your calculation.

Choosing a diluent

Bacteriostatic water — sterile water containing 0.9% benzyl alcohol — is the standard diluent for multi-use research vials because the preservative suppresses microbial growth after the stopper is first punctured. Sterile water for injection contains no preservative and is appropriate only for single-use preparations.

Some sequences need a solubility aid. Acidic peptides often dissolve better with a trace of dilute acetic acid; basic peptides may prefer slightly alkaline conditions. Always test solubility on a small quantity before committing an entire vial.

The concentration maths

Concentration is simply peptide mass divided by diluent volume. A 10mg vial reconstituted with 2ml of bacteriostatic water gives 5mg/ml, which is 5000mcg/ml. On a standard 100-unit insulin syringe, one unit is 0.01ml, so one unit of that solution holds 50mcg.

Technique

Let the vial reach room temperature before adding diluent so condensation does not form on cold powder. Aim the stream of water against the glass wall rather than directly onto the cake, then swirl gently until dissolved.

Do not shake. Vigorous agitation creates an air–liquid interface that denatures and aggregates peptides, and the resulting foam makes accurate withdrawal difficult. If dissolution is slow, wait rather than agitating harder.

After reconstitution

Label the vial with the compound, concentration and reconstitution date. Refrigerate it, protect it from light, and keep records of how long it has been in solution — solution-state stability is a fraction of the lyophilized shelf life.

Frequently asked

Can I reconstitute with tap or distilled water?
No. Neither is sterile and neither is intended for laboratory reconstitution. Use bacteriostatic water for multi-use vials, or sterile water for single-use preparations.
Why can't I shake the vial?
Shaking creates air–liquid interfaces and shear forces that unfold and aggregate peptide chains, reducing the amount of intact material in solution. Gentle swirling and patience are the correct approach.
