The control centre

The arcuate nucleus of the hypothalamus contains two opposing neuron populations. AgRP/NPY neurons drive food-seeking; POMC neurons suppress it. Circulating hormones tune the balance between them minute by minute and season by season.

Because the arcuate nucleus sits next to a region with a permeable blood–brain barrier, it can sample circulating signals directly — which is why peripheral hormones influence a central circuit so effectively.

Short-term signals

Ghrelin rises before meals and stimulates AgRP neurons. GLP-1, PYY, CCK and amylin rise after eating and push the balance the other way, slowing gastric emptying and signalling satiety through vagal and brainstem routes.

These are minute-to-hour signals. They shape individual meals rather than long-term body composition.

Long-term signals

Leptin, secreted by adipose tissue in rough proportion to fat mass, is the classical long-term adiposity signal. Insulin acts similarly in the central nervous system.

The complication documented across the literature is that these systems defend against loss far more vigorously than against gain. Reduced energy availability lowers leptin, raises ghrelin and reduces energy expenditure — a coordinated response that is central to why weight loss is metabolically resisted.

Where research peptides fit

Incretin and amylin analogues are studied because they engage these native circuits rather than bypassing them. That is also why they are studied as separate mechanisms that can be combined — different receptors, different arms of the same regulatory system.
