What Is a Peptide? A Plain-Language Primer for Researchers
Peptides sit between single amino acids and full proteins. Here is how they are defined, how they are built, and why chain length changes everything about handling and stability.

The definition, without the jargon
A peptide is a short chain of amino acids joined by amide bonds, usually called peptide bonds. There is no universal cut-off that separates a peptide from a protein, but most laboratory literature treats chains of roughly two to fifty residues as peptides and anything longer as a protein.
That size matters more than it sounds. Short chains are small enough to be synthesised on a resin in a controlled sequence, purified by chromatography, and characterised precisely by mass. Larger proteins usually have to be expressed in living cells and are far harder to define lot to lot.
How the chain is written
Sequences are written from the N-terminus (the free amino end) to the C-terminus (the free acid end), using three-letter or one-letter codes for each residue. A sequence such as Gly-His-Lys tells you the exact order of residues, which in turn determines the folding behaviour, charge, and solubility of the finished molecule.
Terminal modifications are common and are part of the identity of the compound. Amidation of the C-terminus, acetylation of the N-terminus, or the attachment of a fatty acid chain all change how the peptide behaves in solution, and all of them should be reflected on the certificate of analysis.
Why chain length drives handling
Short peptides are more forgiving of temperature swings but often more sensitive to oxidation at exposed residues such as methionine, cysteine, and tryptophan. Longer or lipidated sequences tend to be more prone to aggregation once reconstituted.
This is why lyophilized powder is the standard shipping and storage format across the industry. Removing water removes the medium in which hydrolysis and deamidation occur, so a properly freeze-dried, sealed vial held cold is far more stable than the same material in solution.
What defines quality in practice
Two numbers describe a research peptide lot: purity and net peptide content. Purity is the proportion of peptide-related material that is the intended sequence, measured chromatographically. Net peptide content is how much of the total vial mass is actually peptide rather than counter-ions and residual water.
